Event Summary:AbstractA/Prof. Alice Wong, School of Biological Sciences, University of Hong Kong, China.Ovarian cancer is an extremely lethal disease. It is the fifth leading cause of cancer death in women and is the most lethal gynecological malignancy. However, despite its clinical importance, the factors that regulate the development and progression of ovarian cancer are among the least understood of all major human malignancies. The surface of the human ovary is covered with a single layer of mesothelial cells, the ovarian surface epithelium (OSE). During each reproductive cycle, the OSE takes part in the cyclical ovulatory rupture and repair. In neoplastic progression, unlike carcinomas in other tissues, new epithelial features appear and become increasingly prominent and stable, while the mesenchymal characteristics of OSE diminish. Ovarian cancer also exhibits a distinctive pattern of metastasis. In contrast to many other types of cancer that metastasize through lymphatics and/or hematogenous routes, ovarian cancer metastasizes by peritoneal dissemination, which relies on the ability of cancer cells to detach from the primary tumor, adhere to, and eventually invade through the peritoneum. All these involve dynamic changes in cell-cell adhesion, which is primarily mediated by cell surface receptors known as cadherins. In this seminar, the unique cadherin profiles in ovarian cancer and the roles of these adhesion molecules in promoting disease development, tumor cell progression, and the formation of ascites will be described. How cadherins perform these functions and their link to a variety of signaling pathways, and their potential as new targets in detecting and treating ovarian cancer will be discussed. Event Summary:ABSTRACT It is only in the last decade that the existence of self-renewing cells in the brain has become fully appreciated. As a consequence, classic models of tumor development in the nervous system have invoked dedifferentiation as a requisite for tumor initiation. New concepts have arisen, however, with the continuing study of "adult" neural stem cells in vivo and in vitro. For example, preparation of neurosphere cultures from primary glioma tissue from human tumors and mouse genetic models permits detailed analysis and the hope for a full molecular understanding of these cells and how they compare to normal stem cells. A central question remains the identification of the cell of tumor origin. Our own work using tumor suppressor models of gliomagenesis or neurofibromas leads us to propose that the cell of origin in these tumors is an early progenitor cell or possibly the primary stem cell. I will discuss the nature of these mouse models, recent advances, how we hope to resolve our current working hypothesis, and implications for sporadic human cancer. If indeed stem or progenitor cells give rise to glioma, it becomes important to fully understand their properties and physiological roles.

Title:IMCB Seminar by Prof. Ali Shilatifard, Transcriptional elongation control and human leukemia
 
Event Summary:AbstractProf. Ali Shilatifard, Investigator, Stowers Institute for Medical Research, USA. Chromosomal translocations involving the MLL gene are associated with infant acute lymphoblastic and mixed lineage leukemia. There are a large number of translocation partners of MLL that share very little sequence or seemingly functional similarities, however, their translocations into MLL result in the pathogenesis of leukemia. To define the molecular reason why these translocations result in the pathogenesis of leukemia, we purified several of the commonly occurring MLL chimeras. We have identified super elongation complex (SEC) associated with all chimeras purified. SEC includes several RNA polymerase II elongation factors including ELL, P-TEFb and AFF4. We have demonstrated that AFF4 is required for SEC stability and proper transcription by poised RNA polymerase II in metazoans. Knockdown of AFF4 in leukemic cells shows reduction in MLL chimera target gene expression suggesting that AFF4/SEC could be a key regulator in the pathogenesis of leukemia through many of the MLL partners. Event Summary:Abstract We present a versatile scheme for the sensitive detection of proteinadditionally functionalized with affinity  ‐fragments we demonstrate that it is possible to discriminate targets according to their size (molecular weight) on‐chip.We describe the underlying principles of the DNA oscillation and show how the dynamics of the switching process can be inferred from frequency response measurements. We demonstrate that the binding of proteins or DNA to the capture probes (i) substantially alters the switching amplitude and (ii) changes the oscillation frequency significantly. Using antibodies and antibody‐ and DNA‐targets. It yields quantitative binding data (kinetics & affinity) and permits the differentiation of proteins according to their hydrodynamic drag. For the first time, probe‐target complexes are identified by their “dynamical fingerprints” using electrically modulated, surface‐bound probes. Moreover, the technique features a simple architecture of the bio‐interface, a chip‐compatible format, and does not require the labeling of targets. Electrically switchable DNA layers constitute the sensor’s central functional element. DNA oligomers are chemically tethered to a gold electrode at one end. Their distal ends are modified with fluorescence labels which are used to monitor the conformation/orientation of the DNA relative to the gold substrate in real‐time, taking advantage of energy transfer that quenches the fluorophore emission as it approaches the surface. For protein sensing, the upper end is‐labels or antigens. The DNA molecules are driven to oscillate between “upright” and “lying” conformations by applying AC potentials (0.2 Hz – 200 kHz) to the supporting gold electrode.

Title:IMCB Seminar by Dr. David Brenner, Molecular Pahtogenesis of Hepatic Fibrosis
 
Event Summary:AbstractThe pattern recognition receptor Toll-like receptor (TLR) 4 plays a central role in innate immune responses. In hepatic fibrosis, portal level of the TLR4 ligand lipopolysaccharide (LPS) is elevated increased intestinal permeability. Both Kupffer cells, the resident macrophages in the liver, and hepatic stellate cells (HSCs), which play a crucial role in hepatic fibrosis, express TLR4. However, the precise contribution of TLR4 in hepatic fibrosis is unknown. Aim: (1) To define the role of TLR4 in hepatic fibrosis and hepatocellular cancer (2) To identify the source of TLR4 ligand during hepatic fibrosis (3) To determine the cellular target of TLR4 ligand in the liver (4) To characterize molecular mechanisms by which TLR4 promotes HSC activation and fibrogenesis. Methods: Hepatic fibrosis was induced by bile duct ligation (BDL) of TLR4-intact (C3H/HeOuJ), TLR4-mutant (C3H/HeJ), MyD88-/- and TRIF-/- mice. To deplete intestinal micro flora, we treated the mice with an antibiotics cocktail (Ampicillin, Metronidazole, Neomycin and Vancomycin) for 4 weeks prior to BDL. TLR4-chimeric mice were generated by Cl2MDP injection, irradiation and bone marrow (BM) transplantation of TLR4-mutant BM into TLR4-intact mice and vice versa. Hepatocellular cancer was induced in TLR4-intact and TLR4 mutant mice by a single injection of DEN. We evaluated hepatic fibrosis by sirius red staining, measurement of hydroxyproline content and profibrogenic gene expression (collagen ?1(I), ?SMA, TGF?1, TIMP1) by quantitative real time PCR (qPCR). Cellular targets of LPS in the liver were analyzed by nuclear translocation of NF-kBp65 by immunofluorescence. To evaluate HSC activation, we measured collagen promoter activity in HSCs isolated from collagen-promoter driven GFP transgenic mice and TGFβ transcriptional activity after stimulation with TGFβ or co-culturing with Kupffer cells (KCs). LPS targets in HSCs were analyzed by microarray and qPCR. Results: Upon BDL, hepatic collagen and hydroxyproline content were less increased in TLR4-mutant mice and antibiotics-treated mice compared with TLR4-intact mice. Combination of Cl2MDP, irradiation and BMT completely reconstituted KCs, but not HSCs, with BM-derived cells, generating TLR4-chimeric mice. Chimeric mice containing TLR4-mutant BM/TLR4-intact HSC had normal fibrosis after BDL, whereas chimeric mice with TLR4-intact BM/TLR4-mutant HSC had reduced fibrosis, indicating HSCs are the main target of TLR4 ligand in hepatic fibrosis. After LPS injection into KC-depleted mice, NF-kB was strongly activated in HSCs. LPS treatment enhanced HSC activity by TGFb- and KC-induced TGF? transcriptional and collagen promoter activity. Microarray analysis revealed 154 LPS-regulated genes in quiescent HSCs including downregulation of the TGF pseudoreceptor BAMBI. BAMBI was downregulated by LPS stimulation in WT-HSCs, but not in MyD88-/- or I?B super-repressor-treated cells. TGFβ transcriptional and collagen promoter activities were upregulated and downregulated in the HSCs overexpressing of dominant negative BAMBI and full length BAMBI, respectively. Upon BDL, profibrogenic gene expression was suppressed in MyD88-/- mice, but not in TRIF-/- mice. TLR4 wild-type mice had more HCCs than TLR4 mutant mice. After 9 months, DEN induced hepatic tumors in wild-type and TLR2-/- but not in TLR4-/- mice. Conclusion: Hepatic fibrosis and HCC is induced by TLR4. HSCs, but not KCs, are the main targets of TLR4 ligands in hepatic fibrosis. TLR4 enhances hepatic fibrosis by downregulating TGF pseudoreceptor BAMBI and enhancing TGFb signaling in HSCs. TLR4 also induces the expression of chemokines that recruit HSCs and Kupffer cells/macrophage to the injured liver. In the fibrotic liver, myofibroblasts and fibrocytes are recruited from the bone marrow, but only constitute a small component of the Type I collagen expressing cells. TLR4 is also required for hepatocarcinogenesis.
